Yes but not shown to other PO type also.

Did you maintain everything in IMG Materials Management/Purchasing/Messages?
Did you assign your new output type in:
1. Assign Form and Output Program for Purchase Order
2. Access Sequences/Define Access Sequence for Purchase Order
3. Message Types/Define Message Types for Purchase Order - in both Maintain Message Types for PO (Did you check there in Output details/General data checkbox Access to conditions?) and Fine-Tuned Control: Purchase Order (New and also Change if needed)?
4. Message Determination Schemas/Define Message Schema for Purchase Order.
Since you mentioned that manually it works problem seems to be in access sequence. Above you mentioned that you maintained MN04 settings for document type only but later on you mentioned that you set it for document type and plant. So my understanding is that:
1. You maintained in MN04 for new output type combination with doc. type and plant.
2. Then you created new PO in ME21N with this document type and plant. Correct?
3. Did you also test in MN04 to set another key combination eg. Doc. type/Purch.Org/Vendor?
